Determination of sophoramine and sophocarpine in rabbit plasma by capillary column gas chromatography and their pharmacokinetics

capillary column gas chromatography was proposed. The recovery of sophoramine from plasma was over 90%. There was a linear relationship between 0.5 and 10μg. The comparative pharmacokinetics of sophoramine and sophocarpine (iv 10 mg/kg) were studied in rabbits. Plasma concentration vs time data were described by an open two compartment model. The pharmacokinetic parameters of sophoramine and sophocarpine were as follows: Vc 0.72 and 1.40 L/kg; CL 17.0 and 28.2 ml/(kg·min-1)；α 0.126 and 0.114 min-1；β 0.0136 and 0.00853 min-1, respectively. The differences in distribution volume, clearance and elimination rate constant of the drugs were significant (p<0.05).
