Pharmacokinetic perfusion model of presystemic metabolism with renal clearance

Abstract

A pharmacokinetic perfusion model of presystemic metabolism with renal clearance (RPPP model) has been developed to descibe the time course of drugs that are subject to preabsorptive, intestinal epithelial, hepatic first-pass metabolism and renal clearance. Equations are derived and solved using statistic moment analysis to estimate the metabolized fractions of the administered dose in each of the three sites. The model is compared with a pharmacokinetic perfusion model of presystemic metabolism without consideration of renal clearance.
