Cytotoxicity of hydroxycamptothecin and four other antineoplastic agents on KB cells

Abstract

The cytotoxicity of l0-hydroxycamptothecip (HCPT), homoharringtonine (HHAR), lycobetaine (LBT), 5-fluorouracil (5-FU), and vincristine (VCR) on KB cell was studied by means of colony formation method and/or trypan blue exclusion method. The results showed that colony formation method was more sensitive to these drugs in comprison with trypan blue method. The order of sensitivity of two methods for these drugs was as follows: VCR>5-FU>HCPT>LBT>HHAR. Colony formation method was particularly suitable for estimation of the cell cycle-specific antineoplastic agents, such as VCR, 5-FU and HCPT. This method seems to be more reliable for determining cell lethality in vitro.
HCPT at 80μg/ml had slight inhibitory effect on incorporation of [3H]TdR into DNA of KB cells. This effect depended on the time and dose. No inhibitory effect was found under the cytotoxic doses. The results indicate that the extent of inhibition of precursor incorporation into DNA synthesis is not directly parallel to its cell killing.
