Don) Hutchins, Fumariaceae, has been used as an anodyne in China. Protopine was isolated from the root. The analgesic and anti-spastic effects of protopine are reported in this paper. The oral LD50 of protopine was 186+/-33 mg/kg in mice. Assayed with the stretching method, the median analgesic dose (ED50) in mice was 27+/-5 mg/kg. The electric stimulation experiment showed that the effect of protopine was weaker than that of morphine. Protopine could inhibit the contraction of duodenum of rabbits at the concentration of 1*10(-5) g/ml. It could antagonize the spasm of isolated ileum of rats induced by acetylcholine or BaCl2.
