Cover: siRNA and shRNA delivery and processing. Delivery of preformed siRNAs is achieved by transient transfection techniques such as lipofection or electroporation. Inside the cytoplasm, a Dicer-containing complex promotes the dissociation of the double-stranded siRNA to 2 single strands and the formation of a catalytically competent RISC. In the case of shRNAs, a vector containing an shRNA expression cassette enters the nucleus via transfection or viral infection. Inside the nucleus, the transcribed shRNAs are exported into the cytoplasm. Dicer-mediated processing towards siRNAs is followed by association with the other RISC components. (page 273-281).
