Cover:Roles of nAChRs in mediating Aβ toxicity. Soluble Aβ1-42accumulation: (1) activates α7-nAChR and induces Ca2+ influx though this receptor and increases Ca2+ release from intracellular Ca2+ pool (ER); (2) combines to intracellular α7-nAChR and favors intracellular plaque formation; (3) activates presynaptic α7-nAChR located on glutamatergic terminal and increases glutamate release, then induces Ca2+ influx via both NMDA receptor and voltage-gated Ca2+ channels; (4) inhibits a4b2-nAChR and causes receptor up-regulation, and these α4β2-nAChR can be activated by endogenous ACh which depolarize membrane potential and in turn leads to the Ca2+ influx via both NMDA receptor and voltage-gated Ca2+ channels. NR1: a7-nAChRs; NR2: non-α7-nAChRs; ER: endoplasmic reticulum (page 1277_1286).
